Abstract
Signalling pathways are critical for cell function, but their dysregulation can lead to diseases such as cancer and inflammation. Our research combines microfluidic live-cell imaging, single-cell analysis and computational modelling to understand how the NF-κB immune pathway processes complex signals. We discovered that single-cell dynamics lead to distinct patterns of gene expression in response to pathogens and cytokines. Introducing a new single-cell proteomics technology called Prox-seq, we simultaneously measured proteins, protein complexes and mRNA in thousands of cells. In human immune cells we identified novel protein complexes such as CD8:CD9 in naive CD8 T cells, and examined protein interactions during toll-like receptor signalling in macrophages. Prox-seq offers a scalable approach to understanding single-cell signalling and can reveal novel protein interactions across a variety of cell types.
